Study of extraction of biologically active substances into fatty base

This diploma thesis deals with macerates of comfrey (Symphytum officinale) in selected fats (food lard, cosmetic lard and almond oil). Selected fats were first characterized by dry matter content, saponification, acid, iodine number, peroxide value. At the same time, total and free fatty acids were determined using GC-FID. Macerates were prepared by extraction of comfrey roots with selected fats. In prepared macerates the change in acid and peroxide value was monitored, as well as the content of selected bioactive compounds. The content of total phytosterol and total carotenoid content was determined by UV-VIS spectrometry, phytosterols and carotenoids were also analysed using HPLC-DAD. Total phenolic content was measured using Folin-Ciocalteu reagent and antioxidant activity by ABTS assay. The measured properties were then compared with industrially produced comfrey ointment. In macerated fats the increase in acid and peroxide value was determined. Furthermore, an increase in the content of total phytosterols and total carotenoids was observed. Using HPLC-DAD the content of -sitosterol and stigmasterol was determined, but carotenoids were not detected. Of the tocopherols, only DL--tocopherol acetate was detected. During maceration, the content of total phenolic compound in fat increased, which caused a change in antioxidant activity.
Experimental study and modelling of the transdermal penetration of active species from gels

This diploma thesis deals with design and experimental study of transdermal transport of pharmaceutically active agents from gel matrices, which contain humic substance in its structure. A model absorption membrane was represented by the skin of pig´s earlobes. The study of the release of active substances and Lignohumte was performed due to the vertical diffusion cells. The amount of released humic substance was characterized by UV-VIS method and the amount of released active agent from gel matrix was determined by HPLC-DAD. The most interesting finding of this diploma thesis was that Lignohumate enhances transdermal transport of active agents and supports their release from gel samples to the particular environment.
Development of a method for determination of antibiotics in milk using high performance liquid chromatography

This diploma thesis focuses on optimization and validation of the method for a mixture of tetracycline antibiotics (oxytetracycline, tetracycline) determination using high performance liquid chromatography with a diode array detector (HPLC–DAD). The aim of this thesis was to determine suitability of the system for the determination of these antibiotics in milk. The theoretical part is focused on individual groups of antibiotics and their mechanism of action, resistances and possibilities of use. Next point of this thesis is legislation, which describes permitted amounts of antibiotics in milk. In this study, the HPLC method and validation parameter is described in detail. The experimental part deals with creation and optimization of the method and its validation. Following validation parameters were verified: system suitability, linearity, method accuracy, limit of detection and quantification, robustness. The results obtained indicate mostly suitable ratio of mobile phase of 0,01 M trifluoracetic acid:methanol:acetonitrile = 80:10:10. The wavelength 355 nm was found as the most feasible for this method after evaluating the result. Determination of robustness showed that the method is robust, and the only significant effect on the robustness was observed in column changes.
The effect of the addition of local wines in the production of wine sausages

The aim of this diploma thesis was to determine the effect of the addition of several types of local wines in the production of wine sausages. Selected wine varieties were characterized using basic and modern analytical methods (liquid chromatography with a diode array detector) and the obtained results were compared with the results of sensory analysis.The sensory analysis was divided into two parts, the first evaluated six non-traditional wine varieties used in the production of wine sausages and the second part evaluated samples with the addition of the traditionally used wine variety Veltlínské zelené, but in three different volumes. Using the Statistica program, the data obtained from the sensory analysis were processed and the mutual correlation was evaluated. Using the gas chromatography method with a flame ionization detector the fatty acid content of a standard sample of wine sausage has been determined. Based on all the determined results, the best wine variety for the production of wine sausages was chosen, it was the blue grape variety André.
Srovnání antioxidačních vlastností a absorbce UV záření u extraktů vybraných bylin, Spiruliny a jejich směsí

This study aimed to compare the antioxidant properties and UV radiation absorption of se-lected herbal extracts, blue-green algae spirulina extract and their mixtures. Extracts were ob-tained by ultrasound assisted extraction. The total polyphenol (TPC) and flavonoid contents (TFC) were determined, and the Trolox equivalent antioxidant capacity (TEAC) was meas-ured. The best antioxidant properties has been showed by C. officinalis individual extract and its mixture with spirulina. Additionally, the UV-absorbing substances at 355 nm were quanti-fied. The extracts from C. officinalis and S. nigra were further analysed for their rutin content by HPLC-DAD. The rutin content in S. nigra and C. officinalis extracts was found to be 1935,97 ± 98,39 μg.g-1 and 233,035 ± 8,839 μg.g-1 of dry weight of flos, respectively. The highest content of phenolic compounds was found in the S. nigra extract, and the highest total antioxidant capacity was determined for the C. officinalis extract. Pearson's correlation re-vealed that the overall antioxidant capacity and UV absorption results were significantly cor-related with the total phenolic compound content only for the elderflower extract. The syner-gistic effect of the herbs used in combination with spirulina was evaluated.
Development of HPLC-DAD-MS/MS method for determination of selected toxins in Amanita phalloides.

(EN) This work has been focused on the development and optimization of analytical methodology, using high performance liquid chromatography with diode array detection (HPLC-DAD) and high performance liquid chromatography with diode array and tandem mass spectrometric detection (HPLC-DAD-MS/MS) for the determination of amatoxins (α- and β-amanitin) and phalotoxins (phallacidin and phalloidin) in the crude extract from cap of Amanita phalloides with the option to later use for clinical purposes. In order to guarantee the reliability of the analytical results the influence of various parameters on the quality of the separation and the determination of toxins was studied. The developed HPLC-DAD method achieves good linearity in the concentrations range 1 - 100 µg/ml, with correlation coefficients higher than 0,997. Limits of detection (LOD) and quantitation (LOQ) were calculated for all the studied toxins with following values: for α-amanitin 0,90 µg/ml (LOD); 2,99 µg/ml (LOQ), β-amanitin 1,07 µg/ml (LOD); 3,56 µg/ml (LOQ), phallacidin 2,17 µg/ml (LOD); 7,26 µg/ml (LOQ) and phalloidin 0,79 µg/ml (LOD); 2,64 µg/ml (LOQ). Impact of post-harvest processing, geographical locality and production year on quality of selected medicinal plants

This study deals with the assessment of the impact of post-harvest treatment, geographical location and production years on 32 selected characteristics of medicinal plants grown in the Czech Republic. Dimethyl sulfoxide herbal extracts characteristics were evaluated by spectroscopic and chromatographic techniques. The results were processed by analysis of variance and multivariate statistical tools. Obtained results clearly proved the successful differentiation of herbal samples by means of canonical discrimination analysis according to post-harvest treatment (100%), geographical origin (82.9%) and production year (96.1%) confirming that studied factors may have significant effect on quality of medicinal plants.
